Development of mAb-loaded 3D-printed (FDM) implantable devices based on PLGA

The main objective of this work was to explore the feasibility print monoclonal antibody (mAb)-loaded implantable systems using fused-deposition modelling (FDM) build complex dosage form designs. Indeed, our knowledge, is first investigation mAb-loaded devices FDM. To make possible, different steps were developed and optimized. A mAb solution stabilized trehalose (TRE), sucrose (SUC), hydroxypropyl-?-cyclodextrin (HP-?-CD), sorbitol or inulin (INU) in order be spray dried (SD). Printable filaments then made poly(lactide-co-glycolide) (PLGA) powder (15% w/w) hot melt extrusion (HME). FDM process optimized these without altering stability. TRE selected associated L-leucine (LEU) increase stability evaluated considering high low molecular weight species levels. mAb-based well-stabilized with excipients during both HME processes. 3D-printed showed sustained-release profiles a burst effect. mAb-binding capacity preserved up 70% following whole fabrication process. These promising results demonstrate that could used produce good stability, affinity mAb.
